The South African midfielder played no part in the Toffees' first Premier League win of the season at Birmingham, Everton prevailing 2-0 after an own goal from Roger Johnson and a late effort from Tim Cahill.
Pienaar, 28, injured his groin during a training session in midweek and faces between two and four weeks on the sidelines.
Moyes said: "He hurt his groin on Wednesday in training and it looks as if he might be out for two to four weeks.
"Two weeks would be good, four would not be. It is a real blow because he played so well at Fulham and is an influential player.
"I'm hoping he can heal quickly and at least we have got the international break coming up."
The Everton boss also revealed that goalscorer Cahill was close to not making the clash at St Andrews with a knee problem.
Cahill, who nodded in Leighton Baines' cross deep into injury-time, has been struggling with the knock and Moyes says he was a doubt.
"There was a doubt about Cahill today. He is still not looking 100 per cent," he added.
"He has not trained enough for me to think everything is absolutely right."
