The summer signing from Nottingham Forest put through his own net against Stoke on Sunday to condemn the Magpies to a 2-1 defeat.
Perch looked distraught at the final whistle, with his mistake having cost Newcastle dear.
Hughton is confident, though, that the 25-year-old can leave that disappointment in the past and come back stronger.
"James is an ever-present in the side and he has to put what has happened behind him," he told the Evening Chronicle.
"Nobody will feel worse about Sunday than James. These things happen, and the only way to put them behind you is by bouncing back.
"It is one mistake and everybody makes them in life, not just football. We know he has strength of character and he will now move on to the next game."
While keen to avoid dwelling on the past, Hughton knows his side could have done without a demoralising defeat on home soil against the Potters.
Their next Premier League fixture takes them to Manchester City, who ended Chelsea's 100 per cent start to the campaign last time out.
Hughton accepts that his side face a tough test at Eastlands on Sunday but, having seen off Everton and Chelsea (in the Carling Cup) in their last two away dates, he insists the Magpies should not be fearful of Roberto Mancini's men.
"We will be going there with belief," he said.
"The players have had a day off and now we will look to get over the disappointment of Stoke very quickly.
"At the moment we are playing in a league that has a lot of different results. And there are going to be shocks every week.
"That's why we are going to Man City with that belief."
